Abstract

Ion channels are transmembrane proteins playing a key role in signal transduction in excitable cells. Mutations in some of human ion channel genes are the cause of various disorders. Furthermore, a number of human ion channels are primary targets of therapeutic drugs used for the treatment of diverse range of human diseases. Despite significant progress over the past 15 years in determining high-resolution structures of some representatives of ion channel family, it is still very difficult to obtain high-resolution structural information for a majority of mammalian ion channels. The ultimate goal of structural modeling methods is to use available structural and experimental data and increase power and accuracy of computational modeling to predict the structure of a channel protein at the atomic level of resolution. Successful pursuit of this goal will be useful for our understanding of ion channel function and modulation on structural level and rational design of drugs targeting ion channels.
