Construction of a cDNA library is a highly sophisticated technology that is used in molecular biology studies. The quality and integrity of the cDNA library is directly related to the success or failure of cDNAs that are of interest to investigators. 1 4 In order to obtain a good library, the person working on the library should have a strong molecular biology background and extensive laboratory experience in molecular biology. Fortunately, there are many methods that have been well developed, and multiple commercial kits are available for cDNA cloning. This chapter describes the detailed strategies used in cDNA synthesis, construction and screening of cDNA libraries, and in the isolation of putative clones.
